Why is my Windows 2000 server accessing the 3.5" disk drive every 5 minutes?

A. If the File Replication Service (FRS) is running on your server, a bug in the service can cause the machine to incorrectly include the 3.5" drive in the drive table and periodically access the drive. Domain controllers (DCs) use the service to replicate information in the Sysvol share, and the Distributed File System (DFS) uses the service on replicated DFS shares.

Microsoft will provide a fix for this problem with Win2K Service Pack 3 (SP3). If you can't wait for the new service pack, you can consult Microsoft for a Win2K Post-SP2 hotfix, as described in the Microsoft article "Changes to the File Replication Service."
